Clyde’s Calcium Assay features:

Ratiometric Dyes

Clyde’s CellOPTIQ® system uses ratiometric dyes to measure calcium

Wavelength Analysis

Analyses the ratio between the two wavelengths

Robust Readout

Obtains measurements unaffected by movement artefact or variations in cell layer thickness

Superior Accuracy

Produces data that is more accurate, with significantly reduced signal-to-noise

Simultaneous Measurements

CellOPTIQ® measures simultaneous voltage and calcium at 10,000 times per second provides detailed information about their interaction in arrhythmic action potentials where EADs are present. Sequential measurement cannot provide this information

Example Data - Verapamil

Verapamil was used as an example of mixed action compound.  Low concentrations of Verapamil (30nM), an antiarrhythmic drug with known mixed hERG and L-type Ca2+ channel activity, had no effect on APD (APD75 = 1.5 ± 0.7%) (Panel A), whereas the amplitude of Ca2+ transient was significantly reduced (45 ± 8%, p<0.05) (Panel B). This further highlights the need for measuring more than one parameter when testing compounds with mixed ion channel activity.
